I'm wondering how close is it to Nana Plaza? And would this be an issue if I stay there with my missus.

It is just about across the street from Nana Plaza. I don't have any personal experience with this hotel, but I did indeed hear that it had been renovated recently (and with good reason - not much had been done in terms of upkeep in the preceding 20 years). Here's the Web site Grand Inn Hotel. Make of it what you will. It is located on Soi 3, Bangkok's "Arab Street." However, the Grand Inn is very close to the corner of Suk Road, so you aren't really in hostile territory.

I'm not sure how to interpret the second question. Are you asking: 1) Would your wife suffer any inconvenience or embarassment by staying in a hotel so close to NEP; or 2) Would it be easy for you to sneak out to NEP? The answer to 1) is "no" and to 2), "yes."

Short answer: The Grand Inn Hotel is OK (according to BKK sources), but unless you are determined to get a hotel as close as possible to NEP, there are better options in that price range.
